Design Notes

The sequence is taken from a patent application, and the conditions for expressing the phenotype (benzalacetone concentration, toxicity concerns, culturing specifications) are found in the paper by the same authors cited below in the References section. This enzyme catalyzes only the last step in the raspberry ketone biosynthesis pathway. Therefore, to observe the desired product, the substrate has to be supplied as well as NADPH. The reaction is summarized on the cited MetaCyc page. Running BALST against the amino acid sequence returns various reductase like enzymes of similar function. It also identifies a putative conserved motif in all of them - the NADP binding site that takes one of two forms: GXGXXG and G[GA]XGXXG The BLAST search also suggests that this enzyme belongs to the family of phenylcoumaran benzylic ether reductase (PCBER) like, atypical (a) SDRs.
